The US Food and Drug Administration (FDA) has approved a new medication called Orzeyful (oveporexton) from Takeda, designed specifically to treat Narcolepsy Type 1 by addressing the condition’s underlying cause. Unlike existing treatments that only help manage symptoms like sleepiness or muscle weakness, this drug works by restoring a missing signal in the brain that people with Narcolepsy Type 1 naturally lack. Currently, about 120,000 people in the US are living with this rare sleep disorder. Dr. Emmanuel Mignot, a leading researcher involved in the drug’s development, explained that previous treatments worked by masking symptoms, such as using stimulants to stay awake or sedatives to improve sleep. However, Orzeyful directly activates the brain receptors that the body’s own natural signals, called orexin, would normally stimulate. This approach helps reduce excessive daytime sleepiness and sudden muscle weakness (known as cataplexy) while improving other related symptoms. The approval is based on results from two major studies, FirstLight and RadiantLight, which showed that the medication significantly reduced these symptoms and improved overall quality of life for patients. Julie Kim, CEO of Takeda, described this approval as a ‘new chapter’ for the Narcolepsy Type 1 community, as it introduces a completely new type of treatment that could change how the disease is managed. The medication is taken by mouth and represents the first drug in its class to directly target the root cause of the condition. Meanwhile, another pharmaceutical company, Eli Lilly, is also investing in this area. In April, they agreed to acquire Centessa Pharmaceuticals for $6.3 billion to develop similar treatments for Narcolepsy Type 1, Narcolepsy Type 2, and a related sleep disorder called idiopathic hypersomnia. This move follows recent leadership changes at Eli Lilly, including the appointment of Dr. Carole Ho as President of Lilly Neuroscience and Dr. Daniel Skovronsky as Chief Scientific and Product Officer, signaling a strong focus on advancing treatments for neurological and sleep-related conditions.
