A new experimental drug called darizmetinib, developed by a company called HepaRegeniX, has received a special designation from the U.S. Food and Drug Administration (FDA). This designation, called Orphan Drug Designation, is given to treatments for rare conditions that affect fewer than 200,000 people in the U.S. It helps support the development of these drugs by providing benefits like tax credits, fee exemptions, and guidance from the FDA. The drug is being tested to help prevent a serious complication called post-hepatectomy liver failure, which can happen after surgery to remove part of the liver. This complication can make recovery difficult and, in severe cases, can be life-threatening. The drug works by helping the liver regenerate faster and more effectively after surgery. Linda Greenbaum, the Chief Medical Officer of HepaRegeniX, explained that post-hepatectomy liver failure is a serious issue that can limit a patient’s ability to recover after major liver surgery. Darizmetinib is designed to enhance the liver’s natural ability to heal and regain function when the remaining liver tissue may not be able to regenerate enough on its own. Elias Papatheodorou, the Chief Executive Officer of HepaRegeniX, said that receiving this FDA designation is a significant step forward for the company and supports the ongoing development of the drug. This designation also strengthens their collaboration with the FDA as they prepare to test darizmetinib in later-stage clinical trials. The drug’s development follows the completion of the first phase of a small early-stage study, called a phase Ib/IIa trial. This part of the study showed that the drug was well-tolerated and had favorable properties in terms of how the body processes it in patients who underwent minor liver surgery. The next phase of the study, which will test the drug in patients undergoing major liver surgery, is expected to begin in the third quarter of 2026. If approved, darizmetinib will receive seven years of market exclusivity in the U.S. for treating post-hepatectomy liver failure, meaning no other similar drug can be sold for this condition during that time.
