This condition affects about 46% of people with Crohn\u2019s disease and currently has no approved medications to treat it. The company has worked closely with the U.S. Food and Drug Administration (<a href=\"https:\/\/www.fda.gov\/\" data-internallinksmanager029f6b8e52c=\"8\" title=\"fda\">FDA<\/a>) to plan this study and expects to start giving the drug to participants in the second half of 2026.<\/p>\n<p>Fibrostenosing Crohn\u2019s disease causes scar tissue buildup in the intestines, which can block the digestive tract. This can lead to problems like pain after eating, trouble absorbing nutrients, malnutrition, and the need for surgery. The NOV-ERA study aims to see if ontunisertib, taken as a pill, can help improve these blockages.<\/p>\n<p>The trial has already received important approvals from regulatory agencies in the U.S., Canada, and other countries, including the European Union and Asia Pacific. The study will involve up to 320 adults with FSCD who have a confirmed blockage in their intestines that prevents a scope from passing through during an endoscopy. Participants will be randomly assigned to receive one of three doses of ontunisertib or a placebo (a dummy pill) twice a day for 52 weeks. Before starting treatment, there will be a 6-week screening period to ensure they are suitable for the trial.<\/p>\n<p>The main goal of the study is to check how many patients have their blockages cleared after 24 weeks of treatment, as measured by an endoscopic scoring system called SES-CD. The trial will also look at other important outcomes, such as changes seen in MRI scans, patient symptoms reported in questionnaires, and how long it takes for FSCD-related problems to occur.
